Contents |
The course aims to provide students with knowledge of the main methods and technologies associated with biochemistry in the context of biotechnological research and in diagnostic and therapeutic applications. The course also includes a part of practical exercises on related topics (see (LABORATORIO PER LE BIOTECNOLOGIE BIOCHIMICHE PER SAGGI MOLECOLARI APPLICATI). |

Educational objectives |
At the end of the course the student will have acquired the basic theoretical knowledge of biochemical methods necessary to work effectively in a laboratory where relevant techniques are used. |
Prerequisites |
Basic knowledge of organic chemistry and biochemistry is required |
Teaching methods |
Frontal lectures. The course is organized to provide the theoretical contents that will then be addressed in the laboratory activities, which are described in the associated teaching (LABORATORY FOR BIOCHEMICAL BIOTHENOLOGIES FOR APPLIED MOLECULAR ASSAYS). |

Il programma del corso comprende esercitazioni pratiche riguardanti la preparazione di soluzioni tampone di interesse biochimico, con l’acquisizione di competenze riguardanti calcoli stechiometrici, diluzioni e percentuali; ii) l’utilizzo di sistemi tampone di interesse biologico e la misurazione del pH; l’estrazione di acidi nucleici e proteine mediante la separazione di fase; l’estrazione di sostanze lipidiche |

Selection of biological samples ( DNA, RNA and Proteins) and quality control analysis. PCR: Theoretical bases and set-up of PCR reactions. Database: nucleotide and amino acid sequences. Real-Time PCR: Theoretical bases, Fluorogenic probes, endogenous housekeeping genes. DNA sequencing and mutational analysis : Sanger method , NGS (Next Generation Sequencing) and Clinical applications. |

Extended program |
The course program includes practical exercises regarding the preparation of buffer solutions of biochemical interest, with the acquisition of skills regarding stoichiometric calculations, dilutions and percentages; ii) the use of buffer systems of biological interest and the measurement of pH; preparation of protein extracts or nucleic acids Determination of proteins and nucleic acids. Enzyme assays. PCR. ELISA assays. Use of specialized instrumentation for the determination of proteins and nucleic acids. |
