- Contents
- Preparation and microscope observation of plant and animal materials. Preparation of microbial cultures. Viable plate count.
- Reference texts
- Teaching material will be provided during the practical laboratory classes.
- Educational objectives
- The main aim of the teaching is to provide students with the bases needed to use the microscope, fundamental instrument for the biologist. Further, they will acquire basic techniques for preparing biological specimen (bacteria, plants, animals).
The main knowledges acquired will be:
parts of the optical microscope and stereomicroscope,
how to use the optical microscope and stereomicroscope,
protocols to prepare fresh biological material and cytological and histological slides from plant and animal organisms; protocols for bacterial cultures.
The main skills will be:
how to use the optical microscope and stereomicroscope,
application of the protocols above mentioned. - Prerequisites
- In order to ensure that the attendance in the practical laboratory classes will be fruitful, it is useful that the students have passed the Cytology and Histology, Botany, Zoology and Microbiology exams.
- Teaching methods
- The course is organized into practical laboratory classes at the Educational Laboratory. Students will be divided into groups (maximum 25 students per group) and they will attend to 8 lab practices of 3 hours each. Lab practices will be tutored, and will also be held in collaboration with Cytology and Histology, Zoology and Microbiology teachers. Attendance is mandatory.
- Other information
- None
- Learning verification modality
- There is not a final examination. However, in order to acquire the "2 cfu" (corresponding to 24 hours), the student will have to attend at least 18 hours (75%) of practical labs. Therefore, a maximum of two absences will be allowed, but not in the same discipline. Frequency will be verified by roll-call and signature take-over.
- Extended program
- Microscopy techniques. Optical and fluorescence microscope; stereomicroscope. Preparation and observation of fresh biological material: microscope examination of zooplankton and phytoplankton, and plant tissues. Use of the stereomicroscope to observe macroscopic Invertebrates. Use of the plant and animal keys. Preparation and observation of cytological and histological slides from plant and animal organisms. Microbiological techniques for the cultivation and isolation of microorganism in the laboratory. Preparation of microbiological culture media. Methods for the isolation of bacterial pure cultures. Procedure for bacteria enumeration by viable plate count.
