Educational objectives Al termine del corso lo studente sarà in grado di:

Comprendere i principi biochimici dei principali saggi molecolari.

Progettare un saggio molecolare in funzione dell’obiettivo applicativo.

Utilizzare correttamente strumentazione di laboratorio per analisi molecolari.

Analizzare criticamente dati sperimentali e redigere un report tecnico.

Applicare criteri di qualità, riproducibilità e controllo (GLP).

Operare in sicurezza in laboratorio biotecnologic

Classificazione dei saggi molecolari biochimici; Saggi qualitativi vs quantitativi; Concetto di target molecolare;
Parametri analitici (accuratezza, precisione, sensibilità); ELISA: principi e varianti;
Saggi enzimatici accoppiati; Rivelazione colorimetrica, fluorimetrica, chemiluminescente; Applicazioni in ambito clinico e industriale;
Cenni su: PCR convenzionale e qPCR; Sonde fluorescenti;
Quantificazione assoluta e relativa; Contaminazioni e controlli; Cenni su: Controllo qualità e GLP
Tracciabilità e gestione del dato Applicazioni in: Diagnostica molecolare, Sicurezza alimentare, Monitoraggio ambientale, Controllo qualità industriale
Attività di laboratorio:
– Tecniche di base e quantificazione proteica:
allineamento su tecniche di base (Uso corretto di micropipette e bilance analitiche; preparazione soluzioni tampone; Curva standard (Bradford )
Analisi spettrofotometrica; Elaborazione dati e costruzione curva di calibrazione
-Saggio immunoenzimatico (ELISA):
Preparazione piastra; Incubazioni e lavaggi; Rivelazione colorimetrica; Lettura spettrofotometrica
Calcolo concentrazione campioni incogniti

Contents Selection of biological samples ( DNA, RNA and Proteins) and quality control analysis. PCR: Theoretical bases and set-up of PCR reactions. Database: nucleotide and amino acid sequences. Real-Time PCR: Theoretical bases, Fluorogenic probes, endogenous housekeeping genes. DNA sequencing and mutational analysis : Sanger method , NGS (Next Generation Sequencing) and Clinical applications.

Educational objectives The main aim of this teaching is to provide students with the theoretical principles that are the basis of the most common techniques of molecular biology, in order to identify possible applications in diagnosis.
Prerequisites The course requires basic knowledge of Biochemistry, Cell Biology and Genetics.

Contents The course aims to provide students with knowledge of the main methods and technologies associated with biochemistry in the context of biotechnological research and in diagnostic and therapeutic applications. Educational objectives At the end of the course the student will have acquired the theoretical and practical knowledge of biochemical methods necessary to work effectively in a laboratory where relevant techniques are used.
Prerequisites Basic knowledge of Organic Chemistry and Biochemistry
Teaching methods l practical lessons in the teaching laboratory relating to the topics that are provided in the course of BIOCHEMICAL BIOTECHNOLOGIES FOR MOLECULAR ASSAYS.

Learning verification modality The course provides basic laboratory skills aimed at improving skills in the health and environmental sectors.
Extended program The course program includes practical exercises regarding the preparation of buffer solutions of biochemical interest, with the acquisition of skills regarding stoichiometric calculations, dilutions and percentages; ii) the use of buffer systems of biological interest and the measurement of pH; preparation of protein extracts or nucleic acids Determination of proteins and nucleic acids. Enzyme assays. PCR. ELISA assays. Use of specialized instrumentation for the determination of proteins and nucleic acids.
